A HPLC machine is a device that is able to separate and analyse the compounds in microscopic samples. The machine works by passing a dissolved mixture, called the mobile phase, through a column containing a series of chemicals with which it will react in one way or another. This is called the stationary phase and last is the retention phase where the sample is eluted from the column.
Gas Chromatography
Gas Chromatography is another method of analyzing compounds and is used where analytes can be vaporized without degrading their properties. Where the mobile phase in HPLC is a liquid (water or inert solution), the mobile phase in Gas chromatography is an inert gas like helium, argon or nitrogen. The stationary phase is a narrow tube called a column where the separated elements of the gas will flow at different rates according to its specific properties. As the separated chemicals leave the column, the gases are identified electronically and the retention time recorded.
There are other forms of chromatography and compound analysers on the market but for all of them, automation is of increasing importance. As well as the efficiency and improved safety issues discussed earlier there is also the benefit of accountability. Tests performed on an automated system are precisely reproducible and this is a crucial advantage when the need to validate analysis can be a legal requirement.
Autosamplers are available for both GC and HPLC, which automate the introduction of samples to the chromatography process. While these may be purchased as part of a new chromatography system, they are also available separately for older machines. They need not be from the same manufacturer that designed the GC or HPLC as long as compatibility is checked prior to purchase.
While manually introducing a sample to a GC or HPLC system will always be an option, labs today are much more likely to be using some form of autosamplers whether that be a simple auto-injecting device or a more complex system involving xyz or rotating robot technology.
Autosamplers are controlled by digital keypad or by computer. Where PC software is used it may come with the autosampler or be sold separately depending on the manufacturer. They may be able to handle large volumes in high throughput labs or limited to smaller quantities. When buying this type of product, it is best to look at what your volume requirements are likely to be in the near future rather than limit to your current capacity.
Another thing to consider, especially when buying an autosampler to connect to a currently set up HPLC or GC system is its configurability to dovetail with the equipment already in the laboratory. Some are top loading to minimise footprint while others offer a left or right option to make the most of available space.
